大规模单细胞转录组图谱揭示COVID-19免疫特征

北京大学张泽民等研究人员合作通过大规模单细胞转录组图谱揭示出COVID-19的免疫特征。相关论文于2021年2月3日在线发表在《细胞》杂志上。

研究人员对来自196名COVID-19患者和对照的284个样品进行了单细胞RNA测序,并创建了一个拥有146万个细胞的全面免疫图谱。大型数据集使研究人员能够确定不同的外周免疫亚型变化与COVID-19的年龄、性别、严重性和疾病阶段等不同临床特征有关。SARS-CoV-2 RNA存在于多种上皮和免疫细胞类型中,并伴随病毒阳性细胞内的转录组显著变化。

S100A8/A9的系统性上调主要由外周血中的巨核细胞和单核细胞引起,可能导致重症患者中频繁观察到的细胞因子风暴。这些数据为了解COVID-19的发病机理和制定有效的治疗策略提供了丰富的资源。 

据了解,COVID-19患者的免疫功能异常是影响症状和死亡率的主要因素,但对相关免疫细胞的详细认知尚不完全。

Abstract: Dysfunctional immune response in the COVID-19 patients is a recurrent theme impacting symptoms and mortality, yet the detailed understanding of pertinent immune cells is not complete. We applied single-cell RNA sequencing to 284 samples from 196 COVID-19 patients and controls and created a comprehensive immune landscape with 1.46 million cells. The large dataset enabled us to identify that different peripheral immune subtype changes were associated with distinct clinical features including age, sex, severity, and disease stages of COVID-19. SARS-CoV-2 RNAs were found in diverse epithelial and immune cell types, accompanied by dramatic transcriptomic changes within viral positive cells. Systemic up-regulation of S100A8/A9, mainly by megakaryocytes and monocytes in the peripheral blood, may contribute to the cytokine storms frequently observed in severe patients. Our data provide a rich resource for understanding the pathogenesis and developing effective therapeutic strategies for COVID-19.
