程序性细胞死亡和凋亡研究历史:细胞死亡史上的重要事件

吞噬细胞(Phagocytes)是Ilya (Elie) Metchnikoff(梅契尼科夫首先描述和记录的,他意识到吞噬细胞吞噬了死亡的细胞。到20世纪中叶,研究人员开始探索细胞是如何死亡的,已经认识到细胞死亡是一个生理控制的过程,最常见的死亡模式(“萎缩坏死”,后来的凋亡)受到严格控制,在1990年之前,一些发现阐明了细胞凋亡的机制。21世纪初,认为自噬过程与细胞凋亡有关,可以限制或增加细胞死亡。之后,研究转向了什么代谢或其他机制间接控制细胞死亡正反馈回路的激活或抑制的问题。

 

 推动这一领域发展的是三项科学进步:

技术上的突破:DNA电泳技术可以证明细胞凋亡,因为在坏死细胞中随机降解的DNA会产生涂片,而在细胞凋亡中,DNA在核小体之间以更有序的方式切割。

概念上的突破:认识到一些疾病,特别是癌症,与细胞死亡的异常模式有关。

理论上的突破:认识到细胞死亡的机制,甚至细胞死亡的组成部分,在从线虫到哺乳动物的进化过程中是保守的。
