GAL4/UAS system(more stronger than heat shock promoter,widely used in drosophila)

UAS:upstream activation sequence;GAL4:a protein from yeast,GAL80 also from yeast.

Tubp-GAL80:cell-type specific promoter drive GAL4,cross with UAS-gene a fly,generate transgene

GAL80 is a temperature control expression protein.

Building transgence can be microinject into fly embryo

2.       Mutant/deletion,EMS

Very limited information

a.P-element insertion,but imprecise excision

b.EMS ,toxic

c.X-ray

***deficiency library,>10kb.

3.       RNAi
