1、禁食推动了热量限制饮食对小鼠的代谢、分子和衰老保护作用

  • 热量限制有益处,长时间禁食抗衰老!

热量限制(Calorie restriction,CR)促进不同物种的健康衰老。最近有研究表明,每天禁食一定时间可以改善新陈代谢并且延长寿命。这些发现使啮齿动物CR研究的解释变得复杂,因为动物通常每天只吃一次,并迅速消耗食物,这相当于间接的强制禁食。在本文中,长时间禁食对CR饮食的关键代谢、分子和衰老保护作用是必要的。通过使用一系列饮食方案,研究人员分析了卡路里和禁食的影响,并进一步证明仅禁食就可以重现CR的许多生理和分子效应。本研究为进食时间和进食量如何调节新陈代谢健康、延长寿命提供了新的解释,并证实了每日长时间禁食可能是CR饮食改善代谢和衰老保护的原因,而不仅仅是热量摄入的减少。

如何做到仅进行卡路里限制?

在本研究中,作者为了研究仅卡路里限制或仅禁食限制造成的影响,主要设计了四种饮食进行对比实验:

1、AL组(对照组):随机给予小鼠正常饲料。该饲料kcal百分比分别为:蛋白质24.0%;碳水化合物58%;脂肪18%。总热量为3.2 Kcal/g。

2、Diluted AL组(仅卡路里限制组):对AL组饲料掺入不可消化的纤维素稀释50%。经测定,热量百分比为蛋白质22.9%;碳水化合物60.5%;脂肪16.6%。总热量为1.6 Kcal/g。该饲料等同于在不禁食的情况下限制30%的卡路里摄入。

3、MF.cr组(卡路里限制+不禁食组):饲料与对照组一致,但食物量减少了30%。且为了减少禁食期,并限制暴饮暴食行为,在12小时的黑暗时间里,使用自动喂食器将食物等分成三份,定时投喂。

4、CR组(卡路里限制+禁食组):饲料与对照组一致,但食物量减少了30%。且只在每天早晨喂食一次;等同于延长餐间禁食时间。

其中,第2组为通过变更饲料成分限制卡路里,但不减少食物量以防止fasting禁食造成影响。而第3组则使用直接降低30%饲料的方式降低卡路里,但通过分批次投喂以减少禁食期。而第4组则通过降低饲料量+每日喂食一次。通过降低食物量从而加速消耗,变相达成长时间禁食的效果。

Abstract

Calorie restriction (CR) promotes healthy ageing in diverse species. Recently, it has been shown that fasting for aportion of each day has metabolic benefits and promotes lifespan. Thesefindings complicate the interpretation of rodent CR studies, in which animalstypically eat only once per day and rapidly consume their food, which collaterally imposes fasting. Here we show that a prolonged fast is necessaryfor key metabolic, molecular and gero-protective effects of a CR diet. Using aseries of feeding regimens, we dissect the effects of calories and fasting, andproceed to demonstrate that fasting alone recapitulates many of thephysiological and molecular effects of CR. Our results shed new light on howboth when and how much we eat regulate metabolic health and longevity, anddemonstrate that daily prolonged fasting, and not solely reduced caloricintake, is likely responsible for the metabolic and geroprotective benefits of a CR diet.

2、模拟禁食饮食预防高脂饮食对心脏代谢风险和寿命的影响

由饮食诱导的肥胖是代谢综合征、糖尿病和心血管疾病的主要危险因素。本文中,研究人员发现,在为期2年的时间里,每4周进行一次5天的模拟禁食饮食(fasting-mimicking diet,FMD)可在雌性小鼠中改善高脂肪、高热量饮食(high-fat, high-calorie diet,HFCD)引起的不利影响。研究表明,每月的FMD周期通过减少内脏和皮下脂肪的积累来抑制HFCD介导的肥胖,而不影响瘦肉重量。这种FMD周期可提高心脏血管功能和对心脏毒素的抵抗能力,预防HFCD诱导的高血糖、高胆固醇血症和高瘦素血症,改善受损的葡萄糖和胰岛素耐受性。每月的FMD周期会改变脂肪细胞中与线粒体代谢和生物合成相关的基因表达,以及造成HFCD喂养小鼠中持续的酮体生成,表明了脂肪细胞重编程在预防肥胖中具有重要作用。FMD对肥胖和心脏衰老的影响可以解释其对HFCD依赖性早期死亡的保护作用。

空腹模拟饮食研究现状

FMD(fasting-mimicking diet,空腹模拟饮食)是以植物饲料为基础,限制热量的低糖、低蛋白质和高脂肪的饮食结构,(fast与refed)呈周期性交替进行,以防止或尽量减少瘦肉质量损失。这种饮食方式可以减缓肿瘤进展和不同类型的癌症对化疗的敏感性,同时保护正常细胞免受化疗毒性副作用。该饮食方式由Valter D. Longo在2012年提出。

除了本研究以外,该课题组还开展了许多围绕FMD的研究。

目前研究发现,FMD可以改善与衰老和多种与年龄相关的疾病的一系列代谢物的表达水平,如代谢综合征、糖尿病和心血管疾病。2018年该课题组在Cell杂志上发表了一篇文章“Fasting-mimickingdiet promotes Ngn3-driven β-cell regeneration to reverse diabetes”,介绍了FMD对糖尿病的影响。短期的FMD循环可以减少IGF1(Insulinlike growth factor-1,胰岛素样生长因子1)、胰岛素和瘦素以及通过上调EGR1(Earlygrowth response 1,早期生长应答因子1)和PTEN(Phosphatase and Tensin homolog,磷酸酶和张力蛋白同源物)抑制PI3K-AKT-mTOR信号传导。通过这种方式,FMD可以增加β细胞数量并促进胰岛素分泌和葡萄糖稳态,从而逆转1型和2型糖尿病。

Fasting-mimickingdiet prevents high-fat diet effect on cardiometabolic risk and lifespan

Abstract

Diet-induced obesity is a major risk factor for metabolic syndrome,diabetes and cardiovascular disease. Here, we show that a 5-d fasting-mimicking diet (FMD), administered every 4 weeksfor a period of 2 years, ameliorates the detrimental changes caused byconsumption of a high-fat, high-calorie diet (HFCD) infemale mice. We demonstrate that monthly FMD cycles inhibit HFCD-mediatedobesity by reducing the accumulation of visceral and subcutaneous fat withoutcausing loss of lean body mass. FMD cycles increase cardiac vascularity andfunction and resistance to cardiotoxins, prevent HFCD-dependent hyperglycaemia,hypercholesterolaemia and hyperleptinaemia and ameliorate impaired glucose andinsulin tolerance. The effect of monthly FMD cycles on gene expressionassociated with mitochondrial metabolism and biogenesis in adipocytes and thesustained ketogenesis in HFCD-fed mice indicate a role for fat cellreprogramming in obesity prevention. These effects of an FMD on adiposity andcardiac ageing could explain the protection from HFCD-dependent earlymortality.

PHGDH的经典途径

PHGDH (Phosphoglycerate dehydrogenase,磷酸甘油酸脱氢酶)是一种催化丝氨酸从头合成的关键酶。其主要活性和代谢功能是在SSP(erine synthesis pathway,丝氨酸合成途径)中,与PSAT1(Phosphoserineaminotransferase 1,磷酸丝氨酸转氨酶)和PSP(Phosphoserine phosphatase,磷酸丝氨酸磷酸化酶)一同将糖酵解产物3-PG( 3-phosphoglycerate,3-磷酸甘油酸)转化为丝氨酸。其中,PHGDH作为限速酶,主要负责将3-PG脱氢转化为3-PHP(3-phosphohydroxypyruvic acid,3-磷酸羟基丙酮酸)。该途径产生NADPH和丝氨酸,其中丝氨酸作为磷酸甘油酯、甘油酯、鞘脂和磷脂丝氨酸的前体,在细胞分化、增殖和迁移中起到重要功能。

在本文中,除了PHGDH在丝氨酸合成中的经典活性功能外,还发现了其参与肿瘤调控,维持肿瘤生长的信号通路,因此称该途径为PHGDH的非典型途径。当葡萄糖缺乏时,PHGDH-Ser371被p38 MAPK磷酸化,促使本应当在胞质定位的PHGDH转位到细胞核,增强了其对苹果酸而非3-PG的氧化活性。当PHGDH激活后,将苹果酸脱氢氧化生成草酰乙酸和NADH并释放能量。而通过抑制PHGDH或阻碍PHGDH进入细胞核则逆转了核内的NAD+水平,抑制肿瘤细胞的生长。

The alternative activity of nuclear PHGDH contributes to tumour growth under nutrient stress

Abstract

The multifunctional roles of metabolic enzymes allowfor the integration of multiple signals to precisely transduce external stimuliinto cell fate decisions. Elevation of 3-phosphoglycerate dehydrogenase(PHGDH), the rate-limiting enzyme for de novo serine biosynthesis, is broadlyassociated with human cancer development; although how PHGDH activity isregulated and its implication in tumorigenesis remains unclear. Herewe show that glucose restriction induces the phosphorylation of PHGDH by p38 at Ser371, which promotes the translocation of PHGDH from the cytosol into thenucleus. Concurrently, AMPK phosphorylates PHGDH-Ser55, selectivelyincreasing PHGDH oxidation of malate into oxaloacetate, thus generating NADH.Inthe nucleus, the altered PHGDH activity restricts NAD+ leveland compartmentally repressed NAD+-dependent PARP1 activity forpoly(ADP-ribosyl)ation of c-Jun, thereby leading to impaired c-Juntranscriptional activity linked to cell growth inhibition. Physiologically,nuclear PHGDH sustains tumour growth under nutrient stress, and the levels ofPHGDH-Ser371 and PHGDH-Ser55 phosphorylation correlate with p38 and AMPKactivity, respectively, in clinical human pancreatic cancer specimens. Thesefindings illustrate a previously unidentified nutrient-sensing mechanism withthe critical involvement of a non-canonical metabolic effect of PHGDH andunderscore the functional importance of alternative PHGDH activity intumorigenesis.
