运动重新编程哺乳动物衰老过程中多个干细胞区的炎症图谱

美国斯坦福大学医学院教授Thomas A. Rando团队近期取得重要工作进展,他们研究发现,运动可以重新编程哺乳动物衰老过程中多个干细胞区的炎症图谱。相关研究成果2023年4月19日在线发表于《细胞—干细胞》杂志上。

据介绍,运动具有使衰老动物的干细胞恢复活力和促进组织再生的能力。然而,运动引起的细胞和分子变化尚未在干细胞区室的广泛细胞类型中进行系统研究。

Title: Exercise reprograms the inflammatory landscape of multiple stem cell compartments during mammalian aging

Abstract: Exercise has the ability to rejuvenate stem cells and improve tissue regenerationin aging animals. However, the cellular and molecular changes elicited by exercisehave not been systematically studied across a broad range of cell types in stem cellcompartments. We subjected young and old mice to aerobic exercise and generated asingle-cell transcriptomic atlas of muscle, neural, and hematopoietic stem cells withtheir niche cells and progeny, complemented by whole transcriptome analysis of singlemyofibers. We found that exercise ameliorated the upregulation of a number of inflammatorypathways associated with old age and restored aspects of intercellular communicationmediated by immune cells within these stem cell compartments. Exercise has a profoundimpact on the composition and transcriptomic landscape of circulating and tissue-residentimmune cells. Our study provides a comprehensive view of the coordinated responsesof multiple aged stem cells and niche cells to exercise at the transcriptomic level.
