中国科学家建立新冠病毒感染的转基因小鼠模型

中国医学科学院秦川、中国疾病预防控制中心病毒病预防控制所Guizhen Wu等研究人员合作利用人源血管紧张素转换酶2(hACE2)转基因小鼠评估了新冠病毒(SARS-CoV-2)的致病性。该项研究成果于2020年5月7日在线发表在《自然》杂志上。

由于ACE2是SARS-CoV-2的细胞进入受体,因此研究人员使用了携带人ACE2并感染SARS-CoV-2的转基因小鼠来研究该病毒的致病性。在感染SARS-CoV-2的hACE2小鼠中观察到体重减轻以及病毒在肺中复制。典型的组织病理学是间质性肺炎,并伴有大量巨噬细胞和淋巴细胞浸入肺泡间质,而且在肺泡腔内积聚巨噬细胞。
 
在支气管上皮细胞、巨噬细胞和肺泡上皮中观察到病毒抗原。在SARS-CoV-2感染的野生型小鼠中未发现该现象。值得注意的是,研究人员确认了hACE2小鼠中SARS-CoV-2的致病性。SARS-CoV-2感染的小鼠模型对于评估抗病毒治疗药物和疫苗以及了解COVID-19的发病机理将具有重要价值。

Abstract: Severe acute respiratory syndrome CoV-2 (SARS-CoV-2) caused the corona virus disease 2019 (COVID-19) cases in China and has become a public health emergency of international concern1. Because angiotensin-converting enzyme 2 (ACE2) is the cell entry receptor of SARS-CoV5, we used transgenic mice bearing human ACE2 and infected with SARS-CoV-2 to study the pathogenicity of the virus. Weight loss and virus replication in lung were observed in hACE2 mice infected with SARS-CoV-2. The typical histopathology was interstitial pneumonia with infiltration of significant macrophages and lymphocytes into the alveolar interstitium, and accumulation of macrophages in alveolar cavities. Viral antigens were observed in the bronchial epithelial cells, macrophages and alveolar epithelia. The phenomenon was not found in wild-type mice with SARS-CoV-2 infection. Notably, we have confirmed the pathogenicity of SARS-CoV-2 in hACE2 mice. The mouse model with SARS-CoV-2 infection will be valuable for evaluating antiviral therapeutics and vaccines as well as understanding the pathogenesis of COVID-19.
