张永振团队破译新型冠状病毒基因组

复旦大学张永振团队的最新研究探究了一种与中国呼吸道疾病相关的新冠状病毒。相关论文2020年2月3日在线发表在国际学术期刊《自然》上。

研究人员对一名在华南海鲜市场工作的患者进行了研究,该患者于2019年12月26日入住武汉中心医院并伴有严重的呼吸系统综合症,包括发烧、头晕和咳嗽。对支气管肺泡灌洗液样品进行超基因组RNA测序,研究人员鉴定了一种新型RNA病毒,并命名为WH-Human-1冠状病毒。对该病毒完整基因组(29,903个核苷酸)的系统进化分析表明,该病毒与先前从中国蝙蝠中获得的一组SARS样冠状病毒(冠状病毒属,轮状病毒亚属)最相似(89.1%核苷酸相似性)。这次疫情突出表明,动物身上的病毒可以引起严重的人类疾病。

Title: A new coronavirus associated with human respiratory disease in China

Abstract: Emerging infectious diseases, such as SARS and Zika, present a major threat to public health1–3. Despite intense research efforts, how, when and where new diseases appear are still the source of considerable uncertainly. A severe respiratory disease was recently reported in the city Wuhan, Hubei province, China. Up to 25th of January 2020, at least 1,975 cases have been reported since the first patient was hospitalized on the 12th of December 2019. Epidemiological investigation suggested that the outbreak was associated with a seafood market in Wuhan. We studied one patient who was a worker at the market, and who was admitted to Wuhan Central Hospital on 26th of December 2019 experiencing a severe respiratory syndrome including fever, dizziness and cough. Metagenomic RNA sequencing4 of a bronchoalveolar lavage fluid sample identified a novel RNA virus from the family Coronaviridae, designed here as WH-Human-1 coronavirus. Phylogenetic analysis of the complete viral genome (29,903 nucleotides) revealed that the virus was most closely related (89.1% nucleotide similarity) to a group of SARS-like coronaviruses (genus Betacoronavirus, subgenus Sarbecovirus) previously sampled from bats in China. This outbreak highlights the ongoing capacity of viral spill-over from animals to cause severe disease in humans.
