Cover Story

 The eukaryotic genome isfunctionally and spatially organized within the cell nucleus. How these spatialorganizations regulate genome function remains largely unclear. This review (pages 255-268) provides an overview on cytological observations of subnucleargene positioning, and summarizes recent findings onthe molecular mechanisms of gene positioning and genome function.

  Visualizing and tracking specific DNA sequences in the cell nucleus canprovide critical information on the spatiotemporal regulation of physiologicalprocesses including transcription, replication and repair. DNA fluorescence in situ hybridization(FISH) allowsdetermining the subnuclear localizations of specificDNA sequences infixed cells, while thelacO/lacI system and the m6A tracer technology enablethree-dimensional tracking of DNA elements inliving cells.The obtained information from these imaging assayscan be correlated with functional activities of these DNA elementsand with genome-wide analyses. Together these techniques willyield important advances in our understandings ofnuclear organizations,epigenetics and genome function.
